How will it operate in future: I would imagine it will continue to operate by running trains through tunnels under London for the foreseeable future
Will it be privatised: Not if Ken has any say in the matter
Will it even become a 24 hour service: If anyone can come up with a practical* way to get all the maintenance done while still running trains, then yes I think it will. Until then I can't see it happening. (*a completely impractical way would be to duplicate all the tunnels)
Sorry for being slightly flippant, but I need more time to think about sensible answers!

Gazing into my crystal ball:
Staffing
In the short/ medium term i.e. next 5 years or so, you'll see the removal of virtually all ticket offices. You MAY see some NR 'travel centre' style offices being introduced to solve problems etc.
In the medium/ long term i.e. 10 - 20 years, I'd imagine ticket offices will be a dim and distant memory and I believe you'll see at least one driver-less tube line with "train captains" instead of drivers.
Privatisation
Well the tube already is part privatised at it isn't exactly working. Further privatisation would require a huge political shift and primary legislation. There is little profit from privatising the services as I'm sure the fares would be capped and any saving will come from a reduction in staff - which TFL will do anyway.
24 hours running
Not within the life of the PPP. Would require a particular commitment from a mayoral politican and for that politican to keep to his words. Perhaps you may see a 24hr service to/from Heathrow but no further than Hammersmith/ Earls Court. Can't imagine a regular 24hr service "in a tunnel". It's just about possible for a 24 hour service one night per week, but it would be costly and I'm not sure it would pass any cost:benefit ratio.
