This will probably be more relevant to current or ex LUL/LT employees,what,in your experience, trains spend/spent most time in service? My vote goes to T352 on current Jubilee WTT 8 Saturday service 04:23 SMD until 01:40 NEAS.

Are we allowed to include trains that operate over periods where there are 24 hour running, such as NYE/NYD?

After spending several hours of thought on this(!), you win - I can't think of a train that clocks up so many hours in service.
Not even Train 407 on the Met line (the Chesham shuttle), which starts and ends at the extremes, is as long.

Does running a train in service for so long on a regular basis have a real impact on wear and tear, or does this not create much of an issue? Also are units 'rotated' so that each unit will complete every set working?

I don't think units are rotated; you just get whats available; having particular units doing particular services would be an operational nightmare. On the days I get the same duty two days running, I've always ended up with different units. It also depends on how the service is running, If one train is 'turned short' or reformed, then a particular unit could end up finishing it's traffic day at another depot.

It's down to the DDM/MM at each depot to book the service,he/she will book back requests for maintenance/cleaning,apart from making sure de-icing units are on the right trains they'll just book trains not required at depots on any running number.
