Hello
During times of part closure on the Met at the weekend does the first train always run example being the 05:12 off of Uxbridge to the city?

yes, depending on the line being open.
Unless the Uxbridge branch is closed there will always be trains running from Uxbridge, but possibly only to Harrow/Wembley/Baker Street/Kings Cross.
You always need to Check before travelling if you have to be there for a specific time.
