Which line had the most trains in service in peak hours.How many was it and when?

The Hampstead Tube, running from Charing Cross to Golders Green and Archway, once ran a service of 44 trains per hour.
(And this was with standard flat crossover layouts at all terminal stations!)

My Da when he was Guard over there once told me that they ran 90 second headways with 120 trains in service in the peak. I don't know if that's 100% but I do know that in the fifties you could stand on the platform and as the rear of one train was fading down the tunnel the front of the next was running in.
