Which journey is fastest? (I going to travelling between 20:15 and 21:15 so it's possible to hit a direct Bank-Gallions Reach train) - Bakerloo Line to Baker Street, Jubilee Line to Canning Town, DLR to Prince Regent
- Bakerloo Line to Waterloo, Jubilee Line to Canning Town, DLR to Prince Regent
- Bakerloo Line to Waterloo, Waterloo & City Line to Bank, DLR to Prince Regent
- Piccadilly Line to Green Park, Jubilee Line to Canning Town, DLR to Prince Regent
- Piccadilly Line to Holborn, Central Line to Bank, DLR to Prince Regent

Bakerloo to Waterloo, Jubilee Line to Canning Town, DLR to Prince Regent will likely be quickest, but Picc to Holborn, Central to Bank, DLR to Prince Regent (change at Canning Town if necessary) wont be far behind though. Piccadilly Circus to Bank via Waterloo is only useful of the Central, Circle, District AND Jubilee are all suspended or severely delayed.
DLR trains from Canning Town to Prince Regent can depart from either high or low level platforms, give yourself 2-4 minutes to get to the low level platforms, 1-2 for the high level platforms.

Surprisingly the TfL journey planner suggests via Green Park or Waterloo.
I would go via Holborn and. Bank.

I have a lot of experience of travelling between that bit of Central London and the eastern branches of the DLR. There is little difference in the timings, but via the Jubilee Line is a couple of minutes quicker and so that is the way the planner recommends (it may also have a bias in there to discourage people from adding to the congestion at Bank?).
Generally on the way to Central London I'll take the slightly quicker route and go via the Jubilee. On the way back I'll more often go for a route with fewer changes, particularly as the high level DLR platforms at Canning Town are very exposed and consequently get very cold.

Think you will find that all the Beckton trains are from Stratford after 19:00.
This is why taking the Jubilee is quicker to get to Canning Town.

No, all the Stratford International trains are too Beckton after about then, but Beckton is also served from Tower Gateway all day.

Think you will find that all the Beckton trains are from Stratford after 19:00 It isn't correct. Beckton branch overview:Beckton branch has Tower Gateway-trains every 10 minutes and Stratford International-trains every 10 minutes during off-peak. Beckton branch has Tower Gateway-trains every 8 minutes and no Stratford International-train during peak. Some trains run from Bank, Lewisham & Woolwich Arsenal to Gallions Reach (typical between 10:00 and 11:00 and again after about 20:30)

I would take the Bakerloo to Waterloo, then Jubilee to Canning Town, changing there for the DLR, as the interchange at Green Park between the Piccadilly/Victoria Lines and the Jubilee Line involves a very long walk which I try to always avoid.

However, the Jubilee line to Canning Town is still needed to get the connection.

You didn't mention the day of the week, but as you raised the question on a Saturday you should note that the Drain doesn't run that late on Saturdays (or at all on Sundays), so option 3 doesn't look a good choice. (It closes at about 2145 on weekdays)
